Occurrence of iris yellow spot virus from onion crops in Egypt

Abstract: Iris yellow spot virus (IYSV) is a infects onion bulb and seed crops in many countries including Egypt. Results of the mechanical inoculation reveled that, small chlorotic lesions and systemic necrosis were observed on both Nicotiana benthamiana and Datura stramonium after 10 days, while there were no symptoms were appeared on the onion plant. The viral biological transmission with Thrips tabaci was highly reported to be efficiently for virus transmitted. Our results confirmed the presentence of viruslike part… Show more

“…In the protective assay, the upper-right halves of the leaves were treated with H. tuberculatum WPE 24 h before mechanical viral inoculation. In contrast, the upper-left halves of the leaves were inoculated with TMV only without any treatment [ 54 , 55 ]. Mock leaves treated with a mixture of equal volumes of DMSO, sterile distilled H 2 O, and phosphate buffer with carborundum were used as the controls.…”

“…Under insect-proof greenhouse conditions, tomato seeds were surface sterilized and cultivated in plastic pots (20 cm in diameter) filled with sterilized soil (clay: sand, 1:1). At 28th days post seedlings, tomato seedlings were transplanted into new pots, and one week later, two upper true leaves of each tomato plant were dusted with carborundum and mechanically inoculated with 1 mL of semi-purified TMV as previously described [43,44]. The experiment was carried out in five treatments, each treatment was comprised of five replicates, and each pot contained three tomato plants.…”

“…1 and Table 1 ). The amino acid sequences of N protein from different IYSV isolates have high similarity, and thus the IYSV N protein is relatively conserved [ 12 , 31 , 32 ]. With these antibodies, we developed the TAS-ELISA and MIA methods for detecting IYSV.…”

“…IYSV was firstly recorded in onion crops in Idaho, USA in 1989 [ 4 ] and then in the Netherlands in 1992, where it was characterized to be a new, distinct orthotospovirus from Iris hollandica [ 5 ]. Subsequently, IYSV was found to occur in [ 6 ] Brazil [ 7 ], Australia [ 8 ], Japan [ 9 ], Chile [ 10 ], Spain [ 11 ], Guatemala [ 12 ], Peru [ 13 ], India [ 14 ], and Egypt [ 15 ].…”
