Exposure of isolated spinach thylakoids to high intensity illumination (photoinhibition) results in the well-characterized impairment of Photosystem II electron transport, followed by degradation of the D1 reaction centre protein. In the present study we demonstrate that this process is accompanied by singlet oxygen production. Singlet oxygen was detected by EPR spectroscopy, following the formation of stable nitroxide radicals from the trapping of singlet oxygen with a sterically hindered amine TEMP (2,2,6,6-tetramethylpiperidine). There was no detectable singlet oxygen production during anaerob photoinhibition or in the presence of sodium-azide. Comparing the kinetics of the loss of PS II function and D1 protein with that of singlet oxygen trapping suggests that singlet oxygen itself or its radical product initiates the degradation of D1.

Singlet oxygen ( 1 O 2 ) is a reactive oxygen species that can function as a stress signal in plant leaves leading to programmed cell death. In microalgae, 1 O 2 -induced transcriptomic changes result in acclimation to 1 O 2 . Here, using a chlorophyll b-less Arabidopsis thaliana mutant (chlorina1 [ch1]), we show that this phenomenon can also occur in vascular plants. The ch1 mutant is highly photosensitive due to a selective increase in the release of 1 O 2 by photosystem II. Under photooxidative stress conditions, the gene expression profile of ch1 mutant leaves very much resembled the gene responses to 1 O 2 reported in the Arabidopsis mutant flu. Preexposure of ch1 plants to moderately elevated light intensities eliminated photooxidative damage without suppressing 1 O 2 formation, indicating acclimation to 1 O 2 . Substantial differences in gene expression were observed between acclimation and high-light stress: A number of transcription factors were selectively induced by acclimation, and contrasting effects were observed for the jasmonate pathway. Jasmonate biosynthesis was strongly induced in ch1 mutant plants under high-light stress and was noticeably repressed under acclimation conditions, suggesting the involvement of this hormone in 1 O 2 -induced cell death. This was confirmed by the decreased tolerance to photooxidative damage of jasmonatetreated ch1 plants and by the increased tolerance of the jasmonate-deficient mutant delayed-dehiscence2.

A variety of natural and synthetic compounds can act as effective photosensitizers in biological systems, including psoralens, flavins, porphyrins, acridines, phenothiazines, xanthenes, quinones, polyenes, haloaromatics and inorganic ions. These, in the presence of light and oxygen, inactivate and modify the integrity of important biomolecules and cellular structures, leading to cell death, mutation and other special function losses [70,118,119]. Many photosensitizers can act as mediators of photodynamic and non‐photodynamic reactions, depending on the experimental conditions. Although a great variety of photosensitized reactions may occur with different sensitizers and substrates, they can be classified into two types of reaction mechanisms, that is, Type I (radical) and Type II (singlet oxygen; 1O2) mechanisms. Type I and Type II reactions for important biomolecules have recently been reviewed [27, 119].
