One-step nucleic acid amplification assay is an accurate technique for sentinel lymph node biopsy of breast cancer patients: a meta-analysis

Abstract: Background:To estimate the accuracy of one-step nucleic acid amplification (OSNA) assay as an intra-operative sentinel lymph node biopsy (SLNB) for sentinel lymph node (SLN) metastasis in breast cancer. Methods:PubMed, Cochrane Library and Web of Science databases were searched by two independent reviewers to retrieve literature with per-patient analysis. The deadline was up until December 2016. A meta-analysis was performed using STATA, Meta-Disc, and Revman software. A random-effects model was used and subgro… Show more

“…It can significantly minimize the sampling limitation compared to histological evaluations. Based on the patients and nodes, the good performance of the assay in this study is similar to other published researches and meta-analysis 2023. Thirdly, the construction of existing models was based on postoperative histological information, including tumor grade, multi-focus and vascular invasion, etc., which means that it is impossible to adopt the existing model in the operation to guide the subsequent axillary operation.…”

“…It's interesting to note that, according to our study, also in the other studies positive predictive value is constantly lower than negative predictive value. This finding had already been extensively reported and discussed from Shi et colleagues in a metanalysis on OSNA in breast cancer on 2833 patients [30]. As already discussed, this fact is likely to be related to the lower sensitivity of classic histopathology examination than OSNA.…”

“…This technique allows to quickly isolate, amplify and quantify mRNA encoding for CK-19; a single examination takes 30–40 min, is highly automated, and does not require sterile working conditions: that makes OSNA suitable for intraoperative examination. The excellent accuracy of OSNA in detecting node metastases in breast cancer has been shown in several clinical studies [[26], [27], [28], [29], [30]]. Later, OSNA has been shown effective in detecting lymph node metastases from other types of tumors, such as colorectal carcinoma, endometrial carcinoma, gastric carcinoma, head and neck squamous cell carcinoma and lung carcinoma [[31], [32], [33], [34]].…”

“…Поэтому обнаружение цитокератина 19 в ЛУ является признаком присутствия в нем опухолевых клеток эпителиального происхождения. Метод был разработан для детекции метастазов рака молочной железы и с тех пор успешно применен для детекции метастазов других опухолей эпителиального происхождения, в частности колоректального рака, немелкоклеточного рака легких, рака матки и щитовидной железы [21][22][23][24].…”
