Ontogeny of the Immune Response in the Ovine Lung

Abstract: Perinatal lambs are increasingly appreciated as a model to study respiratory infections of premature and newborn human infants. To explore the relationship between developmental age and immunological competence in the respiratory tract, the basal levels of expression of genes involved in innate and adaptive immune functions in the lung were examined in pre-term lambs (115 days and 130 days), at birth (145 days) and post-partum (15 days and 3 years old). Our results show that innate immune genes (TLRs-3, -4, -7… Show more

“…Messenger RNA expression of Toll-like receptor-4 and -8 (TLR-4 and -8) in the lung, increases throughout gestation but for a sharp drop in TLR-4 mRNA levels in term lambs [71]. TLR-4 is associated with CD14 that recognizes the F protein of RSV [10,80].…”

“…These lambs are not premature; however, innate immune gene responses, cellular differentiation, and responses to RSV by preterm lambs have significant differences from term lambs [71,79]. In contrast, rodents do not survive preterm birth.…”

“…Error bars = SEM. *P , 0.05; ***P , 0. time points during gestation and compared with adult expression levels as described previously (41,42). Expression of LPO and Duox1 was low preterm, at gestational Days 115 and 130, and at birth, whereas expression was markedly increased in adults ( Figures 5A and 5B).…”

“…Expression of SP-A and SP-D increases throughout fetal lung development in lambs. 91,133 RSV infection of lambs enhances expression of SP-A and SP-D and is associated with viral clearance. 44,61,101,102 Drugs, such as ethanol exposure in utero, reduce SP-A production in lungs of newborns and may explain, at least in part, why ethanol consumption by pregnant mothers is a risk factor for severe RSV disease.…”
