Optical high-resolution analysis of rotational movement: testing circular spatial filter velocimetry (CSFV) with rotating biological cells

Abstract: Circular spatial filtering velocimetry (CSFV) was tested during the microscopic registration of the individual rotations of baker's yeast cells. Their frequency-dependent rotation (electrorotation; ER) was induced in rotating electric fields, which were generated in a glass chip chamber with four electrodes (600 μm tip-to-tip distance). The electrodes were driven with sinusoidal quadrature signals of 5 or 8 V PP with frequencies up to 3 MHz. The observed cell rotation was of the order of 1-100 s per revolution… Show more

“…The image quality would need to be further improved in order to acquire spectra with such low magnification. Filters could help improving the contrast . Another possibility could be to use an automatized microscope stage to observe all 39 quadrupoles with a 20X magnification in sequential videos.…”

“…In summary, the search for consistent dielectric object properties will be more promising the greater the number of methods employed. There is hope that the combination of multiple AC-electrokinetic methods may lead to a new kind of DSCS fingerprinting with a higher resolution for cell and particle structures [29,34,35,65]. The author has declared no conflict of interest.…”

“…The measuring chamber for ROT was based on a rectangular glass microchip with four round‐tip electrodes (Fig. A) . ROT spectra were recorded in the 50 kHz to 150 MHz range, as described previously , using a HP 8130A generator (Hewlett Packard, USA).…”

“…The models make it possible to recalculate electric cell properties from the spectra obtained by different AC‐electrokinetic methods. In the field of biology, interesting applications are the determination, screening or the pursuit of changes in membrane capacitance, membrane conductance, and the cytoplasmic properties of single cells . The methods are also applied to analyze the properties of colloids, nanoparticles, and viruses .…”
