The Agrobacterium rhizogenes-mediated transformation procedure was developed by using the hygromycin B phosphotransferase gene (hph) as a selective marker for the oil-producing fungus Umbelopsis isabellina. Different conditions were combined to increase the transformation efficiency. The highest efficiency was obtained by using A. rhizogenes strain R105 and a vector with zygomycete promoter. Southern blot analysis demonstrated that 71% of transformants contained random integrations of T-DNA sequences under optimal conditions. We randomly selected 115 positive transformants resistant to hygromycin to analyze the amount of total fatty acid and gamma-linolenic acid (GLA). Six transformants produced a higher amount of total fatty acids than the wild strain, and one transformant also produced a higher level of GLA than the wild strain in gas chromatography analysis. This is the first report about using A. rhizogenes strain R105 and germinated conidia to transform successfully the recalcitrant zygomycetes and to obtain transformants with a stable phenotype.

Citrus is one of the most important commercial and nutritional fruit crops in the world, hence it needs to be improved to cater to the diverse needs of consumers and crop breeders. Genetic manipulation through conventional techniques in this genus is invariably a difficult task for plant breeders as it poses various biological limitations comprising long juvenile period, high heterozygosity, sexual incompatibility, nucellar polyembryony and large plant size that greatly hinder cultivar improvement. Hence, several attempts were made to improve Citrus sps. by using various in vitro techniques. Citrus sps are widely known for their recalcitrance to transformation and subsequent rooting, but constant research has led to the establishment of improved protocols to ensure the production of uniformly transformed plants, albeit with relatively low efficiency, depending upon the genotype. Genetic modification through Agrobacterium-mediated transformation has emerged as an important tool for introducing agronomically important genes into Citrus sps. Somatic hybridization has been applied to overcome self and cross-incompatibility barriers and generated inter-specific and inter-generic hybrids. Encouraging results have been achieved through transgenics for resistance against viruses and bacteria, thereby augmenting the yield and quality of the fruit. Now, when major transformation and regeneration protocols have sufficiently been standardized for important cultivars, ongoing citrus research focuses mainly on incorporating such genes in citrus genotypes that can combat different biotic and abiotic stresses. This review summarizes the advances made so far in Citrus biotechnology, and suggests some future directions of research in this fruit crop.

Tropical fruits play an important role in the world economy. They make a substantial contribution to human nutrition and are used for medicine, timber, fuel and livestock feed. The major tropical fruit crops are banana, mango, pineapple, papaya and citrus; however, it is estimated that more than 400 species are harvested in the Asia, Pacific and Oceania region. Other regions such as Central and South America are rich in tropical fruit biodiversity. During the last 20 years, biotechnology has been applied to many tropical fruit species. In Australia, micropropagation systems have been developed for banana, pineapple, papaya, ginger, passionfruit and coffee; and good embryogenic protocols have been employed in banana, mango and papaya. Cryopreservation protocols are being developed for many of the major tropical fruit species. Transformation systems have been used to produce disease resistant cultivars of banana and papaya; and, to prevent blackheart, to control flowering and delay ripening of pineapple. The single significant commercial application of transformation has been the Papaya Ringspot resistant papaya in Hawaii. Molecular marker technology has been used to identify genotypes and to confirm intergeneric hybrids between papaya and related Vasconcellea species. Molecular maps have been developed for some species. Genetic diversity within populations and between related wild species has been determined for some fruits. Specific DAF markers have been identified for sex determination in papaya; a SCAR marker has been developed to identify dwarfism in bananas; and, a CAPS marker to identify a PRSV-P resistant gene in highland papaya. The major application in genomics has been the rapid progress in sequencing the papaya genome in Hawaii.
