Oral edible plant vaccine containing hypoallergen of American cockroach major allergen Per a 2 prevents roach-allergic asthma in a murine model

Abstract: BackgroundAmerican cockroaches (Periplaneta americana) are an important indoor allergen source and a major risk factor for exacerbations and poor control of asthma. We previously reported that allergen components from American cockroaches exhibit varying levels of pathogenicity. Sensitization to major American cockroach allergen, Per a 2, correlated with more severe clinical phenotypes among patients with allergic airway diseases.Materials and methodsIn this study, we examined whether oral plant vaccine-encodi… Show more

“…123 In a study from Taiwan, in a mouse model of asthma, oral treatment with Per a 2 expressed in Chinese cabbage, from an hypoallergenic partial clone Per a 2-372, blunt the development of allergen-specific IgE and IL-4 mRNA expression after challenge with intranasal Per a 2, and had preventive effects on airway hyperresponsiveness and inflammation. 124 Group 3 and 4…”

“…The fact that plant-based vaccine formulations will not require antigen purification will be, without a doubt, the main factor that will make them low-cost alternatives [52], which is necessary to provide wide vaccination coverage in developing and low-income countries. Recent promising reports in the viability of formulating oral vaccines include the case of the Hepatitis B virus surface antigen [53] and models of immunotherapy against allergy with promising results [54], among other diseases.…”

“…Plant biotechnology-based vaccines are becoming a reality, even though their progress has been slower than expected. It is particularly true in oral vaccines, having the main drawbacks of poor reproducibility, a question mark in antigen stability, and bioavailability [128,129].…”
