Oral leptin supplementation throughout lactation in rats prevents later metabolic alterations caused by gestational calorie restriction

Abstract: Oral leptin supplementation in physiological doses throughout lactation in rats prevents most of the detrimental effects on energy homeostasis and metabolic alterations in adulthood caused by inadequate fetal nutrition.

“…The increase in sucrose (energy) ingestion may contribute to increase fat deposition, since fructose has been recognized as highly obesogenic ( Bocarsly et al, 2010 ). Fructose (largely as sucrose) is widely present in many Western diets and can induce obesity, including prenatal obesity ( Szostaczuk et al, 2017 ). In rodents, a sucrose-rich diet can rapidly induce a pathologic condition comparable to human metabolic syndrome ( Santuré et al, 2002 ).…”

“…Animal studies indicate that the metabolic/thermogenic phenotype and related characteristics of offspring adipose tissues such as sympathetic innervation are influenced by nutritional conditions in the perinatal period including maternal diet general traits-e.g., total energy intake, [5,6] diet macronutrient composition, [7] and dietary fat quality [8,9] -and leptin supplementation to suckling pups. [10][11][12] The early postnatal period might be particularly important for the programming of the browning potential of WAT, which, in rodents, mainly develops during this stage, whereas BAT develops mostly during late intrauterine life. [13] Studies on the impact of specific micronutrients and food bioactives, such as polyphenols [14][15][16][17] or vitamin A, [18,19] on the metabolic programming of adipose tissue features remain scarce.…”

“…Only in a few particular cases do we know about the mechanism involved and have the specific tools to resolve the problem (i.e., in the case of leptin deficiency, which affects a small number of individuals, obesity can be treated with leptin injections [ 31 ]). Although, there are already some studies showing that the intake of specific nutrients at early life exerts important effects later in adulthood, as has been shown for leptin [ 32 , 33 ], a protein present in breast milk but not in infant formula [ 34 ] and for β-carotene [ 35 ]. Therefore, despite the identification of specific nutrients still being scarce, it is becoming clear that early nutrition constitutes a first line of intervention and that current evidence suggests dietary influence during pregnancy and lactation as main determinants of obesity in offspring in both animal models and humans [ 32 , 36 , 37 , 38 , 39 , 40 , 41 , 42 ].…”

“…The identification of early biomarkers of adult health outcomes is a remarkable interest of obesity research. Such indicators may provide a useful tool to assess susceptibility to obesity in advance and therefore, contribute to delaying its onset and/or counteracting its development [ 32 , 63 , 64 ].…”
