Organization of gangliosides into membrane nanodomains

Abstract: Gangliosides are glycosphingolipids consisting of a ceramide base and a bulky sugar chain that contains one or more sialic acids. This unique structure endows gangliosides with a strong tendency to self-aggregate in solution, as well as in cellular membranes, where they can form nanoscopic assemblies called ganglioside nanodomains. As gangliosides are important biological molecules involved in a number of physiological processes, characterization of their lateral organization in membranes is essential. This re… Show more

“…The most abundant LCB in fungi is phytosphingosine, while in mammals is sphingosine. However, it is possible to find dihydrosphingosine, in both mammals and yeast [4,56]. In yeast, phytosphingosine is 18 or 20 carbon long [57], though C 18 phytosphingosine is more abundant [4].…”

“…Below 20% mol/mol concentrations, GM1 has a known tendency to segregate, by forming clusters [ 39 ]. Clusters of varying sizes were measured, depending on the concentration of GM1 and the lipid composition of the matrix [ 40 ]. It has been reported, for matrices similar to those studied here, containing cholesterol and sphingomyelin, that GM1 nanodomains are in the disordered fluid phase [ 9 , 40 , 41 ].…”

“…GM1 is also known to form nanoclusters in cell membranes and model membranes [ 58 , 59 , 60 ]. There is wide discrepancy in the reported size, composition and phase of these clusters in model systems [ 60 ].…”
