Oridonin enhances the radiosensitivity of lung cancer cells by upregulating Bax and downregulating Bcl‑2

Abstract: Oridonin is an active component of the traditional Chinese herb Rabdosia rubescens. The present study aimed to evaluate the antitumor effects of oridonin on human non-small cell lung cancer (NSCLC) cells and explore whether oridonin could enhance their radiosensitivity. Oridonin was demonstrated to inhibit the proliferation of SPC-A-1 and HCC827 lung cancer cells in a time-and dose-dependent manner, which was detected using the MTT assay. In addition, pretreatment with oridonin for 24 h prior to irradiation wa… Show more

“…Natural product Ori is the major active ingredient of the traditional Chinese medicinal herb Rabdosia rubescens, and has anti-tumor activity [58][59][60][61][62][63][64][65][66][67][68][69][70][71][72] and anti-in ammatory [131]. Ori and its analogue alone or combined with chemotherapy and radiotherapy was reported to effectively kill tumor cells of leukemia, ovarian cancer, lung cancer, esophageal squamous cell carcinoma, osteosarcoma, breast cancer, colorectal cancer and prostate cancer [58][59][60][61][62][63][64][65][66][67][68][69][70][71][72]. As CSCs have been identi ed as the main center of cancer therapeutic resistance [132][133][134], eradicating CSCs is considered as an effective and powerful strategy to improve current anti-cancer therapeutics [132][133][134].…”

To explore whether combination treatment of hyperthermia and Ori could signi cantly enhance the cell-killing effect on NPC cells and stem-like cancer cells, we rst evaluated the effects of the different concentrations of Ori on cell viability.…”

“…ORI also has anti-bacterial [5], anti-inflammation [6], anti-oxidation [7], depressurization [8], anti-inflammation [9][10][11], immune-modulating [12] and analgesia [13] effects (Table 1). In particular, ORI demonstrates a significant inhibitory effect on the proliferation of more than 20 human cancer cell lines, such as common esophageal cancer [14], lung cancer [15], liver cancer [16], prostate cancer [17], breast cancer [18], and colorectal cancer [19], which is attributed to its specific molecular structure. The α-methylene cyclopentanone (enone) on the D-ring is the essential structure of the anti-cancer activity, which can be impaired by ring splitting or methylene saturation [20].…”
