Ornatins: potent glycoprotein IIb‐IIIa antagonists and platelet aggregation inhibitors from the leech Placobdella ornata

Abstract: The purification and characterization of six isoforms of ornatin, potent glycoprotein Ilb-IIIa (GP IIb-IlIa) antagonists and platelet aggregation inhibitors are described. These isoforms were purified from whole leech homogenates of the leech Plucohdella ornata, a North American leech commonly known as the turtle leech, by trichloroacetic acid precipitation, Sephadex G-50 size exclusion chromatography, G P IIb-IIIa affinity chromatography, and CIS reverse-phase HPLC. Each of the five completely sequenced isofo… Show more

“…Most of the RGD motifs are in the N terminus of their sequences, whereas a RGD motif is in the C terminus of tabinhibitin 3 and -4. All the RGD motifs are positioned in a loop bracketed by cysteine residues as found in other platelet aggregation inhibitors such as variabilin (31), decorsin (32), ornatin (33), and snake disintegrins (34 -36); although tabinhibitins do not share similarity with those known platelet aggregation inhibitors. Tabinhibitins have a molecular weight around 25, whereas other RGD-containing platelet aggregation inhibitors have a much smaller molecular mass (around 5 kDa) (19).…”

“…On the other hand, since the interaction of the GPIIb peptides 296306 and 300-3 12, and the GPIIIa sequences 211-222 and 217-231) with fibrinogen can be inhibited by both RGD and gammachain peptides [7,, it is tempting to speculate that these receptor regions, which are highly conserved between different integrins, may be important for recognition of RGD-containing ligands, i.e. fibronectin, vitronectin, von Willebrand factor, and fibrinogen antagonists like disintegrins [32,33], ornatins [34], mambin [35], and decorsin [36].…”
