Orodispersible Polymer Films with the Poorly Water-Soluble Drug, Olanzapine: Hot-Melt Pneumatic Extrusion for Single-Process 3D Printing

Abstract: Amorphous solid dispersions (ASDs) improve the oral delivery of poorly water-soluble drugs. ASDs of olanzapine (OLZ), which have a high melting point and low solubility, are performed using a complicated process. Three-dimensional (3D) printing based on hot-melt pneumatic extrusion (HMPE) is a simplified method for producing ASDs. Unlike general 3D printing, printlet extrusion is possible without the preparation of drug-loaded filaments. By heating powder blends, direct fused deposition modeling (FDM) printing… Show more

“…Over the last two decades, HME has been employed as a novel cost-effective pharmaceutical manufacturing technique of different oral solid-dosage forms. It has been suitably used in the preparation of immediate-release, novel taste-masked and abuse deterrence tablets formulations [28,29], chrono-modulated drug delivery systems [30], for immediate release formulation of ODF [2,3], and ODF containing poorly water soluble and highly polymorphic drugs [31][32][33]. Thus, the combination of HME and additive printing technology has been shown to offer several advantages [6,33]; first, the ability to fabricate immediate-release, modified-release, and other novel drug delivery dosage forms, second, the ease to prepare personalized oral drug delivery products, • Repaglinide bioavailability was enhanced by the new film-forming alginate derivative.…”

“…All of the obtained ODFs were characterized by a uniform drug content, optimal strength, and rapid disintegration time and exhibited good printability. The formulation containing PEO, PVP, and poloxamer 188 indicated the fastest dissolution time and immediate dissolution profiles [77].…”

“…Finally, ROKIT HEALTHCARE's DR. INVIVO system has become an immensely popular tool that was cited in more than 60 publications (see Fig. 6) in a variety of applications, such as additive manufacturing for tissue engineering and regenerative medicine, [119][120][121][122][123][124][125] drug testing, 126 and pharmaceutical [127][128][129] and biomaterials research. [130][131][132] In addition to the low-cost DR. INVIVO 4D2 series, ROKIT HEALTHCARE just released a new high-cost model in 2020, INVIVO 4D6 HEALTHCARE (Suppl.…”
