Orthotopic hepatocellular carcinoma: molecular imaging-monitored intratumoral hyperthermia-enhanced direct oncolytic virotherapy

Abstract: Objective: To validate the feasibility of molecular imaging-monitored intratumoral radiofrequency hyperthermia (RFH) enhanced direct oncolytic virotherapy for hepatocellular carcinoma (HCC). Methods: This study included in vitro experiments using luciferase-labeled rat HCC cells and in vivo validation experiments on rat models with orthotopic HCCs. Both cells and HCCs in four groups (n ¼ 6/group) were treated by: (1) combination therapy of oncolytic virotherapy (T-VEC) plus RFH at 42 C for 30 min; (2) oncolyti… Show more

“…In recent years, immunotherapy, a new treatment strategy, has provided the expected improvement in the treatment response in preclinical animal studies [115], and its application in clinical research has also been growing rapidly and has attracted widespread attention. In this section, we focus on two notable synergistic OVs-mediated treatment options: (1) OVs and immune checkpoint inhibitors (ICIs) and ( 2) OVs combined with CAR-T.…”

“…When high doses of T-VEC are injected intratumorally under the guidance of ultrasound and optical imaging techniques, radiofrequency ablation enhances the therapeutic effect of the G47D oncolytic virus in a preclinical (Hu et al, 2021b) study. This dual application opens up a new path for treating larger hepatocellular carcinomas locally, while local injection of the virus reduces the viral therapy's systemic toxicity (Song et al, 2019).…”

“…They reported that the hyperthermia resulted from the used radiofrequency electrode enhanced the direct oncolytic effect of the virotherapy. [ 104 ] Intratumoral administration has been investigated in a clinical trial involving CAR T‐cells targeting GPC3. In another clinical trial, adenovirus encoding the suicide gene, HSV/Tk, was administered similarly.…”
