Osseoconductivity of a Specific Streptavidin–Biotin–Fibronectin Surface Coating of Biotinylated Titanium Implants – A Rabbit Animal Study

Abstract: Quasicovalent immobilization of biotinylated fibronectin with the streptavidin-biotin-fibronectin system on smooth surface titanium shows a beneficial faster osseous healing in vivo. Besides, an antifouling effect of the streptavidin-biotin coating was proven.

“…The animals were operated under a general anesthetic by intramuscular injections of a combination of a dose of 35 mg/kg ketamine and a dose of 5 mg/kg xylazine. For the experiments, the tibia model via anterior transdermal approach on both sides was chosen as described before . A randomization was conducted prior surgery via computer‐generated list and in each leg, one screw was inserted after non‐underdimensioned drilling with the respective drill as provided by the manufacturer.…”

“…An acceleration and bony ingrowth with an enhanced bone contact to the materials' surface might lead to less loosening of the fixating screws . An increase of survival and success of dental screw‐shaped Ti implants with these properties could be hypothesized as well . Surface modifications of Ti implants are an effective method to enhance osteoblasts' function and subsequent osseointegration.…”

“…A specific material to be tested could replace the cell-bearing glass panel. Alternatively, the glass panel could be coated with surface ligands in different ways [37]. A potential use for evaluation of stem cell differentiation and/or proliferation with fluid shear stress as a mechanical stimulus may be assumed as well.…”

“…This study was based on our group's previous experiments on modified titanium surfaces where various surface coatings were tested (Kämmerer et al. , ; Heller et al. ).…”

“…FN coating of the BSM was chosen because of our centers experience on the superiority of FN coatings on titanium surfaces (Kämmerer et al. ); whereas VEGF coating was chosen due to the strong paracrine effects VEGF has on EPCs and neovasculogenesis. Hence, the objective of this study was to assess and compare the growth (as described by cell count and cell morphology) and function (as described as PCR and ELISA analysis for Endothelial Nitric Oxide Synthase (eNOS) and cytokine‐inducible Endothelial Nitric Oxide Synthase (iNOS)) of endothelial progenitor cells cultured on a covalently bonded fibronectin coating on deproteinized bovine bone (FN‐DBB) and covalently bonded VEGF coating on deproteinized bovine bone (VEGF‐DBB) (test samples), compared to non‐modified deproteinized bovine bone blocks (control sample).…”
