OUT, a Novel Basic Helix-Loop-Helix Transcription Factor with an Id-like Inhibitory Activity

Abstract: Transcription factors belonging to the basic helixloop-helix (bHLH) family are involved in various cell differentiation processes. We report the isolation and functional characterization of a novel bHLH factor, termed OUT. OUT, structurally related to capsulin/epicardin/Pod-1 and ABF-1/musculin/MyoR, is expressed mainly in the adult mouse reproductive organs, such as the ovary, uterus, and testis, and is barely detectable in tissues of developing embryos. Physical association of OUT with the E protein was pred… Show more

“…As shown in Fig. 5C, the wild-type GFP-Id2 protein repressed the MyoD-E47-induced transactivation in a dose-dependent manner (lanes 7-10), as reported in our previous study using the untagged Id2 construct (26). Given that GFP-Id2 is able to shuttle between the nucleus and the cytoplasm, its repression activity is not surprising.…”

“…pE7-TK-luc is a thymidine kinase (TK) firefly luciferase reporter plasmid harboring seven copies of the E-box sequence and is a derivative of pE7-␤A-luc (25). pCMV-MyoD was mentioned in our previous study (26).…”

“…Other examples include some members of the bHLH (47,48), leucine zipper (49), and homeodomain (50) protein families. The way in which the function of Id proteins is integrated at the level of the subnuclear architecture as shown in the present study may also be relevant to these other transcription factor antagonists, in providing a general mechanism through which they can overcome the high DNA-binding free energy of their target proteins.…”

“…Reduction of heterodimer formation between MyoD and E proteins (including E12 and E47) (Massari and Murre, 2000) has been implicated as a mechanism by which various transcription factors inhibit the capacity of MyoD to activate transcription (Narumi et al, 2000;Sun et al, 2001). We therefore tested if Gli1 reduces heterodimer formation between MyoD and E proteins.…”
