OVER-EXPRESSION OF PROLINE-LINKED ANTIOXIDANT PATHWAY AND MODULATION OF PHENOLIC METABOLITES IN LONG LIFE SPAN CLONAL LINE OFORIGANUM VULGAREIN RESPONSE TO ULTRAVIOLET RADIATION

Abstract: Tissue culture‐generated shoot‐based clonal lines of oregano that have a short and long life span during in vitro culture were investigated to determine the role of proline‐linked pentose phosphate pathway in modulating phenolic response following ultraviolet (UV) exposure. This novel reductant cycling pathway for energy and oxidative pentose phosphate pathway (PPP) is more efficient in long life span O‐17 clonal line in response to UV and this supported antioxidant response. UV treatment stimulated rosmarinic… Show more

“…The extract samples were eluted out in an isocratic manner with a mobile phase consisting of 20 mM potassium phosphate (pH 2.5 by phosphoric acid) at a flow rate of 1 mL min −1 and detected at 210 nm. L-Proline (Sigma chemicals, St. Louis, MO) dissolved in the 20 mM potassium phosphate solution was used to calibrate the standard curve (Kwon et al 2009). The amount of proline in the sample was reported as mg of proline per milliliter and converted to mg g −1 FW.…”

“…After initial shoot growth, O-17 line grew hardier and elongated lignified stems with less lateral branches that reduced the overall vigor and leaves required for comparative analysis with O-3. This O-17 line is a long life span line with slower overall growth and higher phenolic content (Kwon et al 2009). …”

“…After 1 month of growth, the leaf tissues of O-3 lines were collected for biochemical analysis. Due to the slower growth of O-17 lines, we were unable to analyze this line for biochemical activities after 30 days, but the basis of long life span has been well documented based on previous studies (Kwon et al 2009) and was the reason for choice to undertake this initial study.…”
