Food security is a major global issue because of the growing population and decreasing land area. Rice (Oryza sativa L.) is the most important staple cereal crop in the world. Application of nitrogen (N) fertilizer has improved crop yield in the world during the past five decades but with considerable negative impacts on the environment. New solutions are therefore urgently needed to simultaneously increase yields while maintaining or preferably decreasing applied N to maximize the nitrogen use efficiency (NUE) of crops. Plant NUE is inherently complex with each step (including N uptake, translocation, assimilation, and remobilization) governed by multiple interacting genetic and environmental factors. Based on the current knowledge, we propose some possible approaches enhancing NUE, by molecular manipulation selecting candidate genes and agricultural integrated management practices for NUE improvement. Developing an integrated research program combining approaches, mainly based on whole-plant physiology, quantitative genetics, forward and reverse genetics, and agronomy approaches to improve NUE, is a major objective in the future.

Nitrogen is essential for plant growth and development. Improving the ability of plants to acquire and assimilate nitrogen more efficiently is a key agronomic parameter that will augment sustainability in agriculture. A transcription factor approach was pursued to address improvement of nitrogen use efficiency in two major commodity crops. To this end, the Zea mays Dof1 (ZmDof1) transcription factor was expressed in both wheat (Triticum aestivum) and sorghum (Sorghum bicolor) either constitutively, UBI4 promoter from sugarcane, or in a tissue specific fashion via the maize rbcS1 promoter. The primary transcription activation target of ZmDof1, phosphoenolpyruvate carboxylase (PEPC), is observed in transgenic wheat events. Expression ZmDof1 under control of the rbcs1 promoter translates to increase in biomass and yield components in wheat. However, constitutive expression of ZmDof1 led to the down-regulation of genes involved in photosynthesis and the functional apparatus of chloroplasts, and an outcome that negatively impacts photosynthesis, height, and biomass in wheat. Similar patterns were also observed in sorghum transgenic events harboring the constitutive expression cassette of ZmDof1. These results indicate that transcription factor strategies to boost agronomic phenotypic outcomes in crops need to consider expression patterns of the genetic elements to be introduced.

Nitrogen (N) could affect storage root growth and development of sweet potato. To manage external N concentration fluctuations, plants have developed a wide range of strategies, such as growth changes and gene expression.• Five sweet potato cultivars were used to analyse the functions of N in regulating storage root growth. Growth responses and physiological indicators were measured to determine the physiological changes regulated by different N concentrations. Expression profiles of related genes were analysed via microarray hybridization data and qRT-PCR analysis to reveal the molecular mechanisms of storage root growth regulated by different N concentrations.• The growth responses and physiological indicators of the five cultivars were changed by N concentration. The root fresh weight of two of the sweet potato cultivars, SS19 and GS87, was higher under low N concentrations compared with the other cultivars. SS19 and GS87 were found to be having greater tolerance to low N concentration. The expression of N metabolism and storage root growth related genes was regulated by N concentration in sweet potato.• These results reveal that N significantly regulated storage root growth. SS19 and GS87were more tolerant to low N concentration and produced greater storage root yield (at 30 days). Furthermore, several N response genes were involved in both N metabolism and storage root growth.
