Overexpression of a Laccase with Dye Decolorization Activity from Bacillus sp. Induced in Escherichia coli

Abstract: Laccases from bacteria have been widely studied in the past 2 decades due to the higher growth rate of bacteria and their excellent thermal and alkaline pH stability. In this study, a novel laccase gene was cloned from Bacillus sp., analyzed, and functionally expressed in Escherichia coli. The laccase was highly induced in the E. coli expression system with a maximum intracellular activity of 16 U mg-1 protein. The optimal temperature and pH of the purified laccase were 40°C and 4.6, respectively, w… Show more

“…In addition, screening for variants of laccase genes in the genome of basidiomycetes, bacteria, and plants, combined with the use of rational directed evolution approaches, has proven to be efficient for enhanced laccase production and activity. [95][96][97][98][99] Native laccases have been isolated from various domains of life, but the culture media used to isolate and routinely maintain cells for laccase screening selects only "culturable" organisms that represent a small fraction of the wealth of recognized organisms with putative laccase sequences as predicted from metagenomic studies. On the other hand, the development of novel media formulations is expensive and time-consuming, and high proportions of organisms from different taxa remain uncultured.…”

“…In general, heterologous expression approaches of laccase genes include plasmid‐borne or chromosomal integration of these genes under the control of native, surrogate, or synthetic promoters. “…4) Several dyes including indigo carmine and orange G were decomposable with extracellularly expressed cotA laccase [52]. Congo red, Coomassie Brilliant Blue R, and crystal violet were decolorized by intracellularly expressed cotA laccase [53]. Malachite Green, Acid Blue 62, and methyl orange were also decolorized by surface-displayed cotA laccase using E. coli.…”
