Overexpression of trans‐Golgi network t‐SNAREs rescues vacuolar trafficking and TGN morphology defects in a putative tethering factor mutant

Abstract: Summary The trans‐Golgi network (TGN) is a major site for sorting of cargo to either the vacuole or apoplast. The TGN‐localized coiled‐coil protein TNO1 is a putative tethering factor that interacts with the TGN t‐SNARE SYP41 and is required for correct localization of the SYP61 t‐SNARE. An Arabidopsis thaliana tno1 mutant is hypersensitive to salt stress and partially mislocalizes vacuolar proteins to the apoplast, indicating a role in vacuolar trafficking. Here, we show that overexpression of SYP41 or SYP61 … Show more

“…Interestingly, the TGN was disrupted and vesicle formation from the Golgi cisternae was affected in a tno1 mutant, and these defects were rescued by overexpression of either the Qa-SNARE SYP41 or the Qc-SNARE SYP61. These results supported the implication of the tethering factor TNO1 in vesicle fusion at the TGN (Yang et al, 2019).…”

“…However, homolog genes in animal cells participate in: (i) the deactivation of Rab3 [14], a homolog of RabD in plants, (ii) interaction with ATG8 in autophagy processes [59,60], (iii) or activation of a RabGTPase of subfamily C, acting as a guanine exchange factor in degradative pathways and macro-autophagy [61]. Intriguingly, a RabD of plants (RABD2A) localizes with proteins associated with salt-stress tolerance capacity, such as EHD1 or SYP41 [62,63]. In a similar way to expression patterns of Rab3GAP1, Rab3GAP2, and RabGAP10 of S. lycopersicum , the autophagy is a mechanism rapidly induced by salt stress [64].…”

“…Surprisingly, there were only three SSTF DEGs in ERY dataset, among 45 genes analyzed in both datasets (Additional file 8: Table S4 and Additional file 12: Data S1). Two of those genes showed similar FC in both cultivars while the expression of the third one, the recently characterized TDR INTERACTING PROTEIN 3 ( TIP3 ) (Yang et al 2019), was more induced by cold stress in ERY than in S.AND cultivar (Additional file 8: Table S4 and Additional file 12: Data S1).…”
