Ovulatory follicular fluid induces sperm phagocytosis by neutrophils, but oviductal fluid around oestrus suppresses its inflammatory effect in the buffalo oviduct in vitro

Abstract: We have recently shown that the conditioned media from bovine oviductal epithelial cell culture suppress sperm phagocytosis by neutrophils, suggesting that the oviduct around oestrus supplies the anti‐inflammatory microenvironment. To investigate the immune response of neutrophils toward the sperm at ovulation in the buffalo oviduct, we examined (a) a detailed distribution of neutrophils in the oviduct in buffaloes, (b) the effect of ovulatory follicular fluid (FF) and oviductal fluid (OF) on sperm phagocytosi… Show more

“…Samples from the intestinal bulb were fixed in Bouin’s solution (Table 1 ). Samples were processed according to the description by 30 as the following: The fixed samples thoroughly washed in 70% ethanol (3 × 24 h) to get rid of the fixative before subsequent processing. The samples were dehydrated in ascending grades of alcohol (80%, 90%, 100I, 100% II), cleared in methyl benzoate, and then embedded in paraffin wax for three hrs.…”

“…The paraffin-embedded sections were stained according to the procedure of Hoff et al (1985) with modifications according to Abdel-Hakeem et al (2019); Abdel-Maksoud et al (2019); Soliman (2019 a ); Soliman et al (2019); Yousef et al (2019); and Mahmoud et al (2020). The sections were then analyzed using a Leitz DM 2500 microscope with an external fluorescent unit (Leica EL 6000).…”

“…Paraffin-embedded sections were stained with Acridine orange stain using a procedure developed by Hoff et al (1985) with modification according to Abdel-Maksoud et al (2019) and Yousef et al (2019). The stained sections were examined under a Letiz model DM 2500 microscope with illumination provided by an external Leica EL 6000 fluorescence unit.…”
