Oxytocin Receptor Down-Regulation Is Not Necessary for Reducing Oxytocin-Induced Prostaglandin F2α Accumulation by Interferon-τ in a Bovine Endometrial Epithelial Cell Line

Abstract: Interferon-tau (IFNtau) is the embryonic signal responsible for pregnancy recognition in ruminants. The primary action of IFNtau is believed to be mediated through inhibition of prostaglandin F(2alpha) (PGF(2alpha)) released from the endometrial epithelial cells in response to oxytocin (OT). Our working hypothesis was that the antiluteolytic effect of IFNtau also involved modulation of PG production downstream of OT receptor (OTR) and/or cyclooxygenase 2 (COX2). There is currently no OT-sensitive endometrial c… Show more

“…The development of conceptus and endometrium requires biochemical communications as well as cellto-cell interactions; therefore, the communication between the conceptus and the uterine epithelium must be studied not only in coculture systems but also in flexible and dynamic cell environments (Krishnaswamy et al 2009). For these reasons, the bovine EECs established in this study could be a valuable tool for studying such changes, as well as for investigating changes in gene expression, as the implantation process proceeds.…”

“…Finding ESR1 present in the bovine endometrial luminal epithelium at cycle days 12-14 (Robinson et al 2001) might resemble a species-specific peculiarity, which could require the direct binding of interferon-regulatory factors on OXTR interferon response elements for preventing estrogen-induced OXTR upregulation in case of pregnancy (Mann et al 1999, Telgmann et al 2003. Only recently, Krishnaswamy et al (2009) have reported that in vitro IFNT was able to reduce OXT-induced PGF 2a without downregulating either OXTR or PTGS2, implying a direct regulatory mechanism of IFNT on OXT has been yet to be unraveled. Clearly, in the presence of IFNT, PGF 2a pulses impairing luteal function are absent.…”

“…In ruminants, endometrial epithelial cells are the primary source of PGF2␣, whereas stromal cells release more PGE2 (6,7). In bovine uterine flushing, PGF2␣ levels are higher than PGE2 even during early pregnancy (8).…”
