P2Y1receptor switches to neurons from glia in juvenile versus neonatal rat cerebellar cortex

Abstract: Background: In the CNS, several P2 receptors for extracellular nucleotides are identified on neurons and glial cells to participate to neuron-neuron, glia-glia and glia-neuron communication.

“…The presence of the P2Y 1 R on axonal terminals and dendrites in the cerebellar cortex and on cell bodies in organotypic slice co-cultures of the dopaminergic (DAergic) system, co-localised with neuronal markers (e.g. with tyrosine hydroxylase (TH), marker for midbrain DAergic neurons) has been described (Amadio et al, 2007;Heine et al, 2007). This particular subtype is also expressed in the distal region of axons and growth cones in primary cultures of embryonic hippocampal neurons.…”

“…Mature granule cells express several types of P2Rs (Hervas et al, 2003; Amadio et al, 2007); however, very little is known about the functional expression of these receptors in early stages of cerebellar development, especially in GCPs. To that end, we measured intracellular calcium transients induced by P2R agonists in Fluo-3-AM loaded GCPs emigrated from 2-day-old explants.…”

“…These authors found that at early stages (postnatal day 7) of cerebellar development the expression of P2Y 1 R was prominent at the interface between migrating granule cells and Bergman glia, while at postnatal day 21, when neuronal migration is completed, P2Y 1 Rs were mainly found in Purkinje cell body ramifications and on synaptic varicosities, and were absent from Bergman glia (Amadio et al, 2007). …”

“…It is worth of emphasizing, however, that in the case of the 7-9-day-old rats the maturation of the auditory pathway is not yet complete, so the characteristics of these more or less ''immature" astrocytes may reflect an intermediate developmental stage. In fact, data about the differential expression of purinergic receptors in juvenile versus neonatal rats have already been published (Amadio et al, 2007), and similar changes may occur in the cases of the muscarinic receptors as well. At any rate, further experiments are required to understand the precise significance of the cholinergic agonist-evoked Ca 2+ transients of the astrocytes in the signal processing activity of the CN.…”
