Pathogenesis of ANCA-associated vasculitides

Abstract: Antineutrophil cytoplasmic autoantibody (ANCA)-associated vasculitides are characterised by necrotising inflammation of small vessels in conjunction with ANCA directed to either proteinase 3 (PR3) or myeloperoxidase (MPO). The aetiopathogenesis of these disorders is still not fully elucidated but clinical as well as in vitro and in vivo experimental data strongly suggest a role for the autoimmune responses to PR3 and MPO in disease development. Clinically, PR3-ANCA are strongly associated with granulomatous va… Show more

“…Among the MPO-AAV patients, MPA accounted for 97%, with 3 patients diagnosed with GPA [PR3-ANCA (-)]; MPO-ANCA may also be present in some of the patients with GPA (Kallenberg, 2011). Previous studies have reported that there was a stronger correlation between PR3-ANCA and the disease activity of AAV (BVAS), while the relationship between MPO-ANCA and BVAS was not consistent with the previous report.…”

“…The leukocyte-enriched cell suspension was aspirated from the Percoll interface. After the erythrocytes had been removed using lysis solution (139 mM NH 4 Cl and 19 mM Tris), cells were washed with HBSS and resuspended in FCS-containing buffer. Cell viability was assessed by trypan blue staining before flow cytometry or cell transfer experiments.…”

“…Briefly, ANCA produced from auto-reactive B cells activate primed neutrophils and activated neutrophils produce reactive oxygen species and release their lytic contents, initiating vasculitis. Even low doses of pro-inflammatory cytokines, including TNF-alpha, interleukin-(IL) 1 and IL-18, may fuel the inflammation by priming neutrophils and increase their surface expression of ANCA antigens [21].…”
