Pathophysiological implications of neurovascular P450 in brain disorders

Abstract: Over the past decades, the significance of cytochrome P450 (CYP) enzymes has expanded beyond their role as peripheral drug metabolizers in the liver and gut. CYP enzymes are also functionally active at the neurovascular interface. CYP expression is modulated by disease states, impacting cellular functions, detoxification, and reactivity to toxic stimuli and brain drug biotransformation. Unveiling the physiological and molecular complexity of brain P450 enzymes will improve our understanding of the mechanisms u… Show more

“…Dopamine and tyramine are metabolized by CYP2D6, and neurosteroid metabolism also occurs via the isozymes of the CYP3A subgroup. 50 , 51 Studying CBD interaction with neurovascular cytochrome P450 enzymes might also offer new mechanisms of action. It could be possible that CBD-mediated CYP2D6 inhibition increases dopamine levels in the brain, which could help to explain the positive CBD effects in addiction/withdrawal scenarios and might support its 5HT (=serotonin) elevating effect in depression.…”

“…− , OH − ) via abnormal uncoupling of the normal metabolic pathways due to hypoxia-specific signals. [75][76][77] Lysyl oxidases (LOXs) Protein-lysine 6-oxidase, also known as LOX, produces H 2 O 2 as a byproduct during crosslinking between cell-matrix protein elastin and collagen using the lysyl tyrosylquinone cofactor. 78 The enzyme is regulated by Hif-1 or Hif-2 and generates ROS and induces metastasis and cell-matrix adhesion via the FAK/Src signaling pathway in both ischemia and glioma.…”

“…Neural CYP enzymes are considered to play important physiological roles in maintaining neural homeostasis through the biosynthesis and degradation of biologically essential endogenous compounds because previous studies demonstrated that CYP enzyme levels in specific neurons may be as high as or higher than those in hepatocytes and CYP expression is typically distributed to specific populations of neurons (Miksys et al, 2000(Miksys et al, , 2003. The characterization of brain and/or neural CYP enzymes and their localization as well as the identification of endogenous substrates and their metabolic endproducts will provide a better understanding of the role of CYP enzymes in brain physiology, development, and diseases (Ghosh et al, 2016;Toselli et al, 2016). It may also be qualitatively and quantitatively different from that in the liver due to differences in expression or to the presence of unique enzymes in the brain and particular neurons (Stamou et al, 2013).…”
