PCR Typing of Genetic Determinants for Metallo-β-Lactamases and Integrases Carried by Gram-Negative Bacteria Isolated in Japan, with Focus on the Class 3 Integron

Abstract: , 587 strains of gram-negative bacterial isolates demonstrating resistance to ceftazidime and a combination of sulbactam and cefoperazone were subjected to a disk diffusion screening test using sodium mercaptoacetic acid; 431 strains (73.4%) appeared to produce metallo-␤-lactamase (MBL). Of these 431 strains, 357 were found by PCR to carry genes for IMP-1 type MBL (bla IMP-1 ), while only 7 and 67 strains carried the IMP-2 gene (bla IMP-2 ) and the VIM-2 gene (bla VIM-2 ), respectively. Neither VIM-1 nor SPM-1… Show more

“…PCR screening was performed for OXA-type carbapenemases (bla , 24 , 51 and -58-like ) and MBLs (bla IMP-1 , IMP-2 and VIP-2-like ) (Shibata et al, 2003;Turton et al, 2006).…”

“…Screening for production of metallo-β-lactamase was performed using the metallo-β-lactamase inhibitor (21). Strains were inoculated onto MullerHinton agar plates where two Kerby-Bauer disks, one containing 30 µg of ceftazidime and one containing 3 mg of sodium mercaptoacetic acid (SMA) (Eiken), 272 M. OHARA ET AL were placed at a distance of 15-20 mm from each other.…”

“…MBL-encoding genes are usually found as gene cassettes in class 1 integrons (Shibata et al, 2003). Typical class 1 integrons contain two conserved segments (CSs), the 59-CS and the 39-CS (Stokes & Hall, 1989;Bennett, 1999).…”
