PDGF-metronidazole-encapsulated nanofibrous functional layers on collagen membrane promote alveolar ridge regeneration

Abstract: This study aimed to develop a functionally graded membrane (FGM) to prevent infection and promote tissue regeneration. Poly( l -lactide-co- d , l -lactide) encapsulating platelet-derived growth factor (PDLLA-PDGF) or metronidazole (PDLLA-MTZ) was electrospun to form a nanofibrous layer on the inner or outer surface of a clinically available collagen membrane, respectively. The membrane was characterized for the morphology, molecule release pr… Show more

“…Bone morphogenetic proteins (BMPs) were widely reported to enhance bone reconstruction in GBR (Shalumon and Jyh-Ping, 2015). Ho et al (2017) incorporated platelet-derived growth factor (PDGF), a potent mitogen, into the nanofibers to facilitate alveolar ridge regeneration.…”

“…Other electrospun based materials include PLGA (Inanç, Arslan, Seker, Elçin, & Elçin, 2009), type I collagen/PCL/nHA (Col/PCL/nHA) (Wu et al, 2014), nHA/PCL (Yang, Both, Yang, Walboomers, & Jansen, 2009), nHA‐coated silk (Yang et al, 2013), and gelatin methacrylate (GelMA)/nHA microgel arrays (Zhao et al, 2016) nanofibrous scaffolds, which are associated with enhanced cells osteogenesis differentiation. Biomimetic nanofiber membrane composed with fish collagen/bioactive glass/chitosan (Col/BG/CTS), electrospun bi‐layered membranes (poly l ‐lactide‐co‐ d,l ‐lactide) encapsulating platelet‐derived growth factor (PDLLA‐PDGF) inner layer and encapsulating metronidazole (PDLLA‐MTZ) outer layer, or one layer was SF/PCL‐PEG‐PCL incorporating nano‐calcium phosphate (SPCA), another layer is PCL membrane), PDLLA/PLGA electrospun membranes coated with nHA, have been shown to enhanced bone formation (Bottino et al, 2012; Higuchi et al, 2019; Ho et al, 2017; Ma et al, 2018; Rakhmatia et al, 2013; Soltani Dehnavi et al, 2018; Türkkan et al, 2017; Zhou, Liu, et al, 2017).…”

“…Clinically available collagen membranes may be functionalized by electrospinning poly-DL-lactic acid (PDLLA) polymers to enhance periodontal regeneration. In fact, membranes functionalized with antibiotics and growth factor coated PDLLA nanofibers can facilitate healing and regenerative processes while reducing the risk of bacterial infection [27].…”
