Performance Characterization of a Switchable Acoustic Resolution and Optical Resolution Photoacoustic Microscopy System

Abstract: Photoacoustic microscopy (PAM) is a scalable bioimaging modality; one can choose low acoustic resolution with deep penetration depth or high optical resolution with shallow imaging depth. High spatial resolution and deep penetration depth is rather difficult to achieve using a single system. Here we report a switchable acoustic resolution and optical resolution photoacoustic microscopy (AR-OR-PAM) system in a single imaging system capable of both high resolution and low resolution on the same sample. Lateral r… Show more

“…In previously reported studies, 12,27 the time taken to complete OR-scan (4.5 mm × 5 mm) area was 50 min (step size of 2 μm in the Y direction and 3 μm in the X direction). AR-PAM will take 8 min to complete same area (step size of 15 μm in the Y direction and 15 μm in the X direction).…”

“…For example, in optical resolution photoacoustic microscopy (OR-PAM), typical imaging depth up to 1 to 2 mm is obtained with lateral resolution of 0.25 to 5 micron, 180,181 and in acoustic resolution microscopy (AR-PAM), 3 to 8 mm imaging depth is obtained with lateral resolution of 40 to 50 micron. 182,183 Most of the PAM systems reported so far have used visible or NIR-I window due to the strong hemoglobin absorption. Recently, OR-PAM was reported for the first time in the second NIR window.…”
