Peroxyoxalate Chemiluminescence Assay of Hydrogen Peroxide and Glucose Using 2,4,6,8-Tetrathiomorpholuiopyrimido[5,4-d]-pyrimidine as a Fluorescent Component

Abstract: Peroxyoxalate chemiluminescence (CL) assay of hydrogen peroxide (H2O2) or glucose was developed by using 2, 4,6,8-tetrathiomorpholinopyrimido[5,4-d]pyrimidine as a fluorescent component and bis(2,4,6-trichlorophenyl)oxalate (TCPO) as an oxalate. Linear relationships between CL intensity and final concentration of H2O2 from 10.8 to 10-4 M were obtained. The detection limit at the ratio of CL intensities for sample and blank (S/ B) of 3 was 10 nM. The precision for five replicate measurements at 10-5 and 10-6 M … Show more

“…Comparing to other analytical techniques such as spectrometry [2], titrimetry [3], and chemiluminescence [4], electrochemical enzyme biosensors [5 -7] based on redox proteins or enzymes modified electrodes possess unique advantages such as simplicity, high selectivity and intrinsic sensitivity. Horseradish peroxidase (HRP), a heme enzyme, is generally used as a representative to investigate the dynamic and thermodynamic mechanism of the enzyme reactions and develop biosensors for the determination of H 2 O 2 or related compounds.…”

“…38,39 By using this combination, sensitive determination of hydrogen peroxide was achieved with an LOD of 10 nM at a signal-toblank ratio of 3. 40 Furthermore, substrates for oxidase such as glucose, uric acid, 41 mono-acetyl polyamines and polyamines 42 could be determined. The method was based on the enzyme reaction to generate hydrogen peroxide, which was performed on-line using an immobilized enzyme column reactor.…”

“…PO-CL assay of hydrogen peroxide or glucose was developed by using TMP and TCPO. 43 a-D-Glucose was transformed to ,3-D-glucose with mutarotase and converted to H2O2 and D-gluconic acid with glucose oxidase, which was detected by using PO-CL reaction. This method was applied to the assay of glucose in human serum.…”
