Persistent Hz-1 Virus Infection in Insect Cells: Evidence for Insertion of Viral DNA into Host Chromosomes and Viral Infection in a Latent Status

Abstract: Persistent/latent viral infections of insect cells are a prominent though poorly understood phenomenon. In this study, the long-term association between the Hz-1 virus and insect host cells, conventionally referred to as persistent viral infection, is described. With the aid of a newly developed fluorescent cell-labeling system, we found that productive viral replication occurs by spontaneous viral reactivation in fewer than 0.2% of persistently infected cell lines over a 5-day period. Once viral reactivation … Show more

“…Cell Lines, Viruses, and Transfection. Sf21 cells, derived from Spodoptera frugiperda , were maintained as monolayer cultures in tissue culture flasks at 28 °C in TNM‐FH medium supplemented with 8% heat‐inactivated fetal bovine serum (3, 22–24). These cells were used for the propagation and infection of wild‐type AcMNPV and recombinant virus.…”

“…Characterization of VLPs by electron microscope. Ultra-structure of cells co-infected with recombinant baculoviruses was collected, fixed, and then visualized by electron microscopy as described in our previous studies [20]. To visualize purified VLPs using transmission electron microscopy with negative staining, an aliquot of 10 ll of the VLP preparation was loaded onto a carbon-coated grid, letting standstill for 5 min.…”

“…Even where there is clear evidence of a disease epizootic, as in many forest pests, both the precise role of disease and its fate within and, more importantly, between epizootics, have yet to be resolved (Myers 1988). One hypothesis is that some pathogens may become 'quiescent', producing hidden or covert infections, and persist within their insect hosts without obvious signs of disease (Longworth & Cunningham 1968;Jurkovicova 1979;Kukan 1999;Lin et al . 1999;Burden et al .…”
