Pharmacokinetics and Pharmacodynamics of Morroniside: A Review

Abstract: Morroniside belongs to a group of iridoid glycosides and is the primary active component of Cornus officinalis Sieb. et Zucc. This species is a rich source of iridoid glycosides and has been used as a traditional Chinese medicinal herbs for centuries. In this review, we discuss the pharmacokinetics and pharmacodynamics of morroniside. Morroniside is quickly absorbed and metabolized and is widely distributed throughout the body. It also exerts various pharmacological effects. Numerous pharmacological studies ha… Show more

“…MR has been documented to possess anti-oxidation, antiinflammation, and anti-apoptosis properties (Gao et al, 2020;Deng et al, 2021;Park et al, 2021). It can exert a neuroprotective role in neurodegenerative diseases by eliminating ROS to enhance the total antioxidant capacity of the rat cortex, thereby preventing nerve cells from H 2 O 2 -induced oxidation injury (Wang et al, 2009;Yao et al, 2009;Zhang et al, 2017c;Li B et al, 2019). Likewise, MR has been proven to protect lung activity against aging by maintaining the cell proliferative state and normal cell morphology and inhibiting apoptosis (Chen et al, 2014).…”

“…Morroniside has been shown to exert many biological effects, such as anti-inflammatory, anti-apoptotic, anti-oxidant, cardioprotective and neuroprotective via several mechanisms and signaling pathways, i.e., STAT3, NF-κB, MAPK and PI3K/Akt pathways and molecules regulating the autophagic flux [ 184 , 185 , 186 , 187 , 188 , 189 ]. It has thus long been used to treat degenerative diseases of the nervous system, to inhibit platelet aggregation, to prevent diabetic angiopathies and renal damage, to treat OA and to reduce bone resorption [ 190 ].…”

“…Several studies have been conducted to determine the concentration of morroniside in biological matrices [9][10][11]. The absolute oral bioavailability of morroniside in rats was calculated to be only 3.6-7.0% [9,12]. The plasma levels of morroniside administered intravenously are much higher than those after oral administration.…”
