Pharmacokinetics and tissue depletion of doxycycline administered at high dosage to broiler chickens via the drinking water

Abstract: The recommended use of doxycycline (DC) to broiler chicken is 100 mg/L via the drinking water and a 7-day withdrawal time (WDT). However, study of a higher dosage is desirable because of the possible increase of antimicrobial resistance and disease spectrum. “…Для вивчення брали чисту плазму курей (попередньо перевірену методом УЕРХ-МС/МС на вміст доксицикліну), збагачували стандартним розчином доксицикліну на рівні 1 і 5 мкг/мл, і послідовно розводили в 5000 разів буфером для розведення зразків, що надається у наборі, для того, щоб отримані розчини, які за вмістом аналіту потрапляли в концентраційний інтервал калібрувальної кривої, і далі проводили аналіз імуноферментним методом згідно з інструкцією. Для навантаження обрали такі концентрації доксицикліну, порядок яких близький до вмісту його в плазмі крові курей-бройлерів при лікуванні їх препаратами на основі доксицикліну (Anadon et al, 1994;Laczay et al, 2001;Wijayanti et al, 2011;Hsiao et al, 2016;Pai-Feng at al., 2016;Yang et al, 2018). Для перевірки правильності та ефективності такого підходу, ці ж зразки було проаналізовано методом УЕРХ-МС/МС, використовуючи калібрувальний графік в інтервалі 0 -10,0 мкг/мл, побудований на матриці чистого зразка плазми крові курчат-бройлерів.…”
