Pharmacokinetics of furosemide in man after intravenous and oral administration. Application of moment analysis

Abstract: Furosemide 40 mg was administered to 8 healthy subjects as an i.v. bolus dose, as 1 tablet in the fasting state, and as 1 tablet and a solution after food intake. The i.v. data gave a total body clearance of 162 +/- 10.8 ml/min and a renal clearance of 117 +/- 11.3 ml/min; the volume of distribution at steady state was 8.3 +/- 0.61. Oral administration gave a bio-availability of the tablet (fasting) of 51%. Food intake slightly reduced the bioavailability, but not to a significant extent. There was no signific… Show more

“…Furosemide strongly binds to plasma proteins (91-99%) (34), particularly to anionic sites on albumin (20). Most intestinal furosemide absorption occurs in the stomach and small intestine (30,58,160). Although the mean availability of oral furosemide is ϳ60%, absorption ranges from 10 -100%.…”

“…Pharmacokinetics of furosemide has been compared between IV and PO administration. 9 However, the effects of furosemide on LAP in dogs with MR have not been well documented in a quantitative manner because there are some difficulties in measuring LAP directly. We have previously reported the 24-hour LAP profiles of and the effect of angiotensin-converting enzyme (ACE) inhibitors on experimentally induced MR dogs with a radio telemetry system.…”

It is well known that furans exhibit pharmacological activity and many of them are in use as pharmaceuticals (Martorana, Share & Zimmerman, 1985;Hammarlund, Odlind & Paalzow, 1985), herbicides (Melhus, 1935), fungicides (Koschucharoff & Harisanowa, 1962), bactericides (Ascoli, Casini, Ferappi & Tubaro, 1967), etc.…”
