Phospholipase C-? and ovarian sex steroids in pig granulosa cells

Abstract: We compared the membrane effects of estradiol, progesterone, and androstenedione in a single experimental model, the ovarian granulosa cells collected from immature Large White sows. We measured changes in cytosolic free calcium concentration ([Ca2+]i) in confluent Fura-2 loaded cells. We used pharmacological tools and polyclonal phospholipase C-beta (PLC-beta) antibodies. Each steroid (0.1 pM to 1 nM) transiently increased intracellular calcium concentration ([Ca2+]i) within 5 sec. They mobilized Ca2+ from th… Show more

“…In normal healthy porcine [35], human [36], and rat GCs [37] as well as SIGCs [25] [39][40][41]. These increases in [Ca 2ϩ ] i are sufficient to allow the transition from G 0 /G 1 to the S phase of the cell cycle [39].…”

“…Our results suggest that estradiol, p,p 0 -DDE, o,p-DDE and methoxychlor act through a pertussis toxin-sensitive G-protein, since pretreatment with the toxin inhibited the [Ca 2þ ] i response to stimulation by these agents. Estradiol and androstenedione act via a pertussis toxin-sensitive G-protein in porcine granulosa cells (Lieberherr et al 1999). On the other hand, the [Ca 2þ ] i response to progesterone and Kepone was not inhibited, suggesting that these two compounds acted through a pertussis toxin-insensitive G-protein.…”

“…In addition, a rapid progesteroneinduced increase in the formation of inositol 1,4,5-trisphosphate (IP 3 ) and 1,2-diacylglycerol (DAG) was observed in these cells (191,251). Similarly, progesterone triggers an immediate and transient peak in [Ca 2ϩ ] i and an induction of IP 3 in porcine granulosa cells (257,272). The mentioned effects in osteoblasts and granulosa cells have been shown to be insensitive to RU 486 and were also triggered by progesterone bound to BSA, although at 100-fold higher concentrations.…”

“…Furthermore, androstenedione, the main intrafollicular androgen, rapidly increased [Ca 2ϩ ] i in human granulosa luteinizing cells (273) and porcine ovarian granulosa cells (257). This increase was dose dependent, with maximum activity at 1 nM androstenedione in pig and 10 nM in human cells.…”
