Photocleavable Polyethylene Glycol for the Light-Regulation of Protein Function

Abstract: PEGylation is commonly employed to enhance the pharmacokinetic properties of proteins, but it can interfere with natural protein function. Protein activity can thus be abrogated through PEGylation, and a controllable means to remove the polyethylene glycol (PEG) group from the protein is desirable. As such, light affords a unique control over biomolecules through the application of photosensitive groups. Herein, we report the synthesis of a photocleavable PEG reagent (PhotoPEG) and its application to the light… Show more

“…The design and fabrication of polymers featuring light‐responsive groups has gained intensive interest in the biomedical field . Light as a spatially and temporally controllable external stimulus can be readily tuned and focused to trigger changes in the structure of polymer chains, and thereby provide “on‐demand” drug delivery . Several methods to synthesize polymeric nanoparticles bearing light‐responsive moieties have been developed by self‐assembly of block copolymers in selective solvents .…”

“…Another example for a photocleavable PEGylation strategy was published by the Deiters group [42]. Using an ortho-nitrobenzyl linked PEG, they showed reversible caging of proteins and peptides, and most importantly the timed regulation of biomolecules activity.…”

“…[1] Such concepts can be realized by different approaches. One of them is the use of cleavable, bifunctional linker units,w hich represents am ethod for triggering the complete decomposition of the polymeric material with external stimuli. Numerous photocleavable polymers, such as polyurethanes, [3] polyacrylates, [4] polyesters, [5] and polyethers, [6] have been reported.
