Phylogenetic Status of Two Undescribed Zygomycete Species from Korea:Actinomucor elegansandMucor minutus

Abstract: During a survey of fungal diversity of the order Mucorales, three zygomycete isolates, CNUFC-YR113-1, CNUFC-KNU16-7, and CNUFC-BS1-1 were isolated from freshwater and soil samples in Korea. The strains were analyzed both morphologically and phylogenetically based on internal transcribed spacer and 28S rDNA gene sequences. Based on their morphology and phylogeny, the CNUFC-YR113-1 and CNUFC-KNU16-7 isolates were identified as Actinomucor elegans, and CNUFC-BS1-1 was identified as Mucor minutus. To the best of o… Show more

“…The columellae of short sporangiophores described here are conical and subglobose, unlike those reported for the extype, which are cylindrical to ellipsoid, some conical to flattened. Nguyen et al (2017) observed the presence of one to three septa below the columellae, unlike our isolate and the ex-type (Schipper 1975), which have no septum below the columellae.…”

“…Variability in nucleotide sequences in the ITS region has been reported by several authors as a critical barcode marker for identifying fungi at the species level, including in the order Mucorales [36,37]. In previous studies, we successfully identified species of Mucorales using this marker [10,22,23,38].…”

“…To date, few studies have reported new and undescribed zygomycetous fungi in Korea [10,23,34,35]. Particularly, species of Backusella and Circinella are rarely found in Korea.…”

“…These samples were transferred in sterile 50 mL conical tubes and stored at 4 °C until examination. Fungi were isolated by a serial dilution plating method as described previously [21], and cultured on potato dextrose agar (PDA: 39 g PDA in 1 L of deionized water; Becton, Dickinson and Co., Sparks, MD, USA) supplemented with streptomycin sulfate 0.5 mg/mL, to suppress bacterial growth. After incubation at 25 °C for 3–7 days, individual hyphal tips of the developing fungal colonies were removed and placed onto PDA medium.…”
