Phylogenetical Relationships between Kejobong Goat and Other Goats Based on Mt-DNA D-loop Sequence Analysis

Abstract: The aim of this study was to investigate phylogenetical relationships between Kejobong goat and Kacang goat as well as Etawah Grade goat using genetic diversity, haplotype, and genetic distance analysis based on D-loop sequences. A total of 76 blood samples belonged to three local goats, namely Kacang goat (KC), Etawah Grade goat (EG), and Kejobong goat (KJ). The DNA genome was extracted based on the manufacturer's standard protocol using gSYNC DNA Mini Kit (Geneaid Biotech Ltd.) for sequence analysis control … Show more

“…A total of 45 Simeulue buffaloes collected from three subdistricts for measurement their body size characteristics. The buffalo samples were grouped into two age categories, 4-6 and 2-4 years with criteria according to Lestari et al (2018). Blood sampling was carried out by collecting about 3 mL of blood samples from buffalo through the jugular vein using a venoject and vacutainer tubes with EDTA and then stored at 4⁰C.…”

“…This similarity of amino acid mutation position perhaps related to the assumption that Kejobong goat was a crossbred of Kacang goat and Etawah Grade goat [ 2 , 3 ], and it was confirmed by Lestari et al . [ 42 ] reporting that by phylogenetic relationship analysis based on D-loop sequence, Kejobong goat was in the same cluster together with Kacang goat and Etawah Grade goat with no clear grouping in each breed in B haplogroup. Meanwhile, as comparator based on other mtDNA genes, Kejobong goat had 11 nucleotide polymorphisms on the basis of 548 bp D-loop sequence that formed 7 haplotypes [ 8 ].…”

“…Another study conducted by Lestari et al . [ 42 ], Kejobong goat had 12 nucleotide polymorphisms of 1191 bp D-loop sequence that formed 11 haplotypes with Pi 0.00143±0.00018.…”

“…The Kejobong goat is known as an indigenous Indonesian breed raised by a semi-intensive animal farming system by local farmers. This goat has been confirmed to be the progeny of a cross between Kacang and Etawah grade goats [1,2]. The Kejobong goat is known not only for its prolific traits but also for its high rate of growth, good carcass composition, and good reproductive performance [3].…”
