Phylogeny of the Yeast Genera Hanseniaspora (Anamorph Kloeckera), Dekkera (Anamorph Brettanomyces), and Eeniella as Inferred from Partial 26S Ribosomal DNA Nucleotide Sequences

Abstract: Partial 26s ribosomal DNA sequences of species assigned to the genera Hanseniaspora, Klueckera, Dekkera, Brettanomyces, and EenieZZu were determined. A phylogenetic analysis of the sequences showed that the genus EenieZZu is derived within the genus Brettanomyces and that the genus Hanseniaspora (anamorph Kloeckera) is not closely related to the genus Dekkera (anamorph Brettanomyces). As a consequence, the name EenieZla is reduced to synonymy with the name Brettanomyces. In addition, our data do not support re… Show more

“…Also, according to our results (Fig. 5), Hanseniaspora, Dekkera and Saccharomyces belong to different groups, in agreement with the findings of other authors using rDNA and rRNA nucleotide sequences (Boekhout et al, 1994 ;Cai et al, 1996 ;Kurtzman, 1994). Within the fourth subgroup, Candida species cluster together.…”

“…This background makes it necessary to have available molecular techniques able to shed light on the taxonomy of these micro-organisms. Recently, molecular techniques involving rRNA and rDNA sequencing have become available for conducting phylogenetic and classification studies (Boekhout et al, 1994 ;Cai et al, 1996 ;James et al, 1997 ;Kurtzman, 1994), and RFLP and RAPD have been applied to identification (Kurtzman, 1994). For example, the sequencing of D1\D2 domains allows the rapid identification of ascomycetous yeast (Fell, 1993).…”

“…In addition, the relatively hydrophobic character of PNAs compared to DNA oligonucleotides makes PNA probes capable of penetrating the hydrophobic cell wall following mild fixation conditions that do not lead to disruption of cell morphology (14). These unique characteristics of PNA have opened new possibilities for molecular diagnostic assays. The D1-D2 region of 26S ribosomal DNA (rDNA) of eucaryotic organisms shows a high degree of species variation and has been used for identification and taxonomy of yeast species (1,8). In this study, 26S rDNA sequence information was used to design species-specific probes targeting the rRNA of D. bruxellensis.…”

“…O nome Brettanomyces é uma alusão ao termo British, devido ao uso de espécies deste gênero na elaboração de cerveja de aroma forte (FRANSON, 2001 CLARK-WALKER, 1986;BOEKHOUT et al, 1994).…”
