Physical mapping of Malacosoma neustria nuclear polyhedrosis virus genome and its modification in Antheraea pernyi cell culture

Abstract: A physical map of the M. neustria nuclear polyidrosis virus (ManeNPV) genome was constructed, the complete order of BamHJ, Kpnl and Pstl restriction enzyme sites was determined t a polyhedrin gene was localized on the map. The viral DNA size was calculated to be about 139 kbp. Restriction endonuclease profiles of the DNA of ManeNPV plaques isolate propagated in A. pernyi cells demonstrated ^persistent heterogeneity*, submolar bands were shown to appear in a digestion pattern of DNA of the first passage virus. … Show more

“…The linearization site was previously delineated on the physical map of the ManeNPV genome with respect to the Bam HI, Kpn I, and Pst I cleavage sites [23] . In the present study, the Bam HI-D fragment of the genome harboring the linearization site was cloned into pBR325.…”

“…An examination of the nucleotide sequence of the ManeNPV Bgl II- Hind III fragment ( Figure S2 ) revealed that the Pst I site, which is located proximal to the linearization site on the ManeNPV physical map [23] , is situated 17 bp upstream of the CE. Based on the comparison of the Pst I site position (17 bp upstream of the CE, in accordance with the sequencing data, and less than 100 bp upstream of the linearization site, in accordance with the ManeNPV physical map, with the CE size (155 bp), it was concluded that the linearization site must be located within the CE.…”

“…The CNE was originally identified at the ends of the linear form of the Malacosoma neustria nucleopolyhedrovirus (ManeNPV) genome (45). The CNE has been demonstrated to act in cis for the BV production of AcMNPV and is suggested to be involved in the replication, recombination, or cleavage of the viral genome (41).…”
