Physicochemical Properties of Chromosomes in Live Cells Characterized by Label-Free Imaging and Fluorescence Correlation Spectroscopy

Abstract: The cell nucleus is a three-dimensional, dynamic organelle that is organized into many subnuclear bodies, such as chromatin and nucleoli. The structure and function of these bodies is maintained by diffusion and interactions between related factors as well as dynamic and structural changes. Recent studies using fluorescent microscopic techniques suggest that protein factors can access and are freely mobile in mitotic chromosomes, despite their densely packed structure. However, the physicochemical properties o… Show more

“…ODT reconstructs 3D RI tomography of a cell sample from multiple 2D hologram images of the sample acquired at various angles (Kim et al, 2018a). ODT measurement of single living cells was performed with a commercial ODT microscope (HT-2H; Tomocube) using a 532 nm laser light source at 37°C in an atmosphere containing 5% CO 2 (Jung et al, 2016;Kim et al, 2018c;2019a;Park et al, 2010). The commercial ODT microscope which is based on Mach-Zehnder interferometry equipped with a digital micromirror device (DMD) to reconstruct the 3D RI tomography of cells.…”

“…Due to its high-resolution, label-free, and quantitative imaging capability, ODT has been recently utilized in various studies, including research in the fields of haematology 28,29 , infectious diseases 30,31 , drug discovery 32 , and cell biology 33 . Analog to X-ray computed tomography, ODT reconstructs the 3-D RI of a microscopic object using a laser beam [34][35][36][37] .…”
