The aim of this research work is to analyze the chemistry and diversity of forest VOCs (volatile organic compounds) and to outline their evidence-based effects on health. This research work was designed as a narrative overview of the scientific literature. Inhaling forest VOCs like limonene and pinene can result in useful antioxidant and anti-inflammatory effects on the airways, and the pharmacological activity of some terpenes absorbed through inhalation may be also beneficial to promote brain functions by decreasing mental fatigue, inducing relaxation, and improving cognitive performance and mood. The tree composition can markedly influence the concentration of specific VOCs in the forest air, which also exhibits cyclic diurnal variations. Moreover, beneficial psychological and physiological effects of visiting a forest cannot be solely attributed to VOC inhalation but are due to a global and integrated stimulation of the five senses, induced by all specific characteristics of the natural environment, with the visual component probably playing a fundamental role in the overall effect. Globally, these findings can have useful implications for individual wellbeing, public health, and landscape design. Further clinical and environmental studies are advised, since the majority of the existing evidence is derived from laboratory findings.

Natural medicines are predominantly used by the majority of the world population as the primary health care (Balick, 1994). Monoterpenes are secondary metabolites of plants and are also widely found in microorganisms. Monoterpenes belong to the category of terpenoids and are also known as isoprenoids (Kohl et al., 2015). Monoterpenes are frequently utilized in many areas like agriculture, cosmetic, food industries, and as general antiseptic. Monoterpenes possess various pharmacological properties like antibacterial, antifungal, antioxidant, anticancer, vasorelaxant, hypotensive, and antispasmodic effects (Tan et al., 2016) (Vieira et al., 2018). They are also used in many medical practices (Ravichandran et al., 2018; Suh et al., 2017). Limonene is a monoterpene that is found in natural fruits like grape fruit (95%), tangerine (94%), orange (91%), mandarine (72%), lemon (65%), and elemi (50%) (González-Mas et al., 2019). D-limonene is the principal active form of limonene (Erasto & Viljoen, 2008). D-limonene is present in citrus essential oils and spices but the most commonly food source of limonene is orange peel oil, which is about 90%-95% D-limonene by weight (Aazza et al., 2011). Limonene is frequently used as a dietary supplement and as a fragrance ingredient for cosmetics products.

Pain and inflammation are complex clinical conditions that are present in a wide variety of disorders. Most drugs used to treat pain and inflammation have potential side effects, which makes it necessary to search for new sources of bioactive molecules. In this paper, we describe the ability of LASSBio-1586, an N-acylhydrazone derivative, to attenuate nociceptive behavior and the inflammatory response in mice. Antinociceptive activity was evaluated through acetic acid-induced writhing and formalin-induced nociception tests. In these experimental models, LASSBio-1586 significantly (p<0.05) reduced nociceptive behavior. Several methods of acute and chronic inflammation induced by different chemical (carrageenan, histamine, croton oil, arachidonic acid) and physical (cotton pellet) agents were used to evaluate the anti-inflammatory effect of LASSBio-1586. LASSBio-1586 exhibited potent anti-inflammatory activity in all tests (p<0.05). Study of the mechanism of action demonstrated the possible involvement of the nitrergic, serotonergic and histamine signaling pathways. In addition, a molecular docking study was performed, indicating that LASSBio-1586 is able to block the COX-2 enzyme, reducing arachidonic acid metabolism and consequently decreasing the production of prostaglandins, which are important inflammatory mediators. In summary, LASSBio-1586 exhibited relevant antinociceptive and anti-inflammatory potential and acted on several targets, making it a candidate for a new multi-target oral anti-inflammatory drug.
