Plant regeneration from suspension cells induced from hypocotyls derived from interspecific cross Alstroemeria pelegrina × A. magenta and transformation with Agrobacterium tumefaciens

Abstract: Embryogenic cell suspension cultures were established using the ovule culture of an interspecific cross, Alstroemeria pelegrina var. rosea × A. magenta. Ovules harvested 14 d after pollination were cultured on Murashige and Skoog (MS) medium without plant growth regulators (PGRs); calli were produced on the hypocotyl surface in germinating zygotic embryos. Suspension cells were induced from the calli by using liquid MS media containing 2,4-dichlorophenoxyacetic acid or 4-amino-3,5,6-trichloropyridine-2-carboxy… Show more

“…1e) and distinguishable perianth shapes are considered to be useful for the analysis of floral zygomorphy. Furthermore, an Agrobacterium-mediated transformation system has now been established in Alstroemeria (Kim et al, 2007;Hoshino et al, 2008). Thus, Alstroemeria will continue to be used in future studies to investigate the role of CYC in the expression of floral zygomorphy.…”

“…Furthermore, molecular mechanisms controlling flower development in this plant species have been studied by analysing class B genes (Hirai et al, 2007) and LEAFY-like gene (Hirai et al, 2012). Additionally, genetic transformation of Alstroemeria has been established by using Agrobacterium tumefaciens (Kim et al, 2007;Hoshino et al, 2008). These research findings will be useful for further analyses of flower development in this plant species.…”

“…Typically, embryo rescue and ovule culture have been effectively used to overcome genetic incompatibility and develop interspecific hybrids in several crops (Morgan, 2004;Kaushal et al, 2005;Eeckhaut et al, 2007;Vlachostergios et al, 2007;Hoshino et al, 2008). Nevertheless, both techniques are not very useful when pro-embryos are used, as they can be damaged during excision due to their very small size.…”
