Plant regeneration of eucalypts from rotating nodule cultures

Abstract: Nodule cultures were induced from shoot tips of aseptically grownEucalyptus botryoides in a vertically rotated incubator. N-(2-chloro-4-pyridyl)-N'-phenylurea (4PU) was a very effective cytokinin for induction of nodules in liquid B5 basal medium supplemented with naphthalene acetic acid and 3% sucrose. The effective concentration of 4PU ranged from 0.2 to 0.5mg/l. The nodule had a very smooth surface and was composed of small meristematic cells outside and large vacuolated cells inside. Shoots were regenerated… Show more

“…Following this, a protuberance formed on the brim of each explant and gradually grew into an intact shoot organ. Similar observations have been reported in Eucalyptus gunnii, Populus, and Eucalyptus grandis (McCown et al 1988;Ito et al 1996;Herve et al 2001). This suggested that the protuberance originated from the stem vascular system, which is consistentwith an earlier report in Populus (Douglas 1984).…”

“…The optimal TDZ concentrations for E. urophylla (Tibok et al, 1995), E. grandisÂurophylla (Cid et al, 1999) and E. globulus (Chriqui et al, 1991;Azmi et al, 1997) were 50, 40 and 20 times higher, respectively, compared to our study. CPPU has been tested in only four other eucalypts (Ito et al, 1996), and was effective at the same concentrations as this study.…”

“…Such structures are reported to resemble compact organogenicmeristemoid tissue commonly formed during cell redifferentiation and shoot regeneration in callus tissues (Ovečka et al 2000). The development of organogenic nodules following a wounding treatment has been reported from stem, leaf and root explants of Chichorium intybus (Pieron et al 1998), while the cytokinin-mediated induction of organogenic nodules has been reported in several species of Eucalyptus (Ito et al 1996) and in Ananas comosus (Teng 1997).…”
