Poly(N‐phenylglycine)‐Based Nanoparticles as Highly Effective and Targeted Near‐Infrared Photothermal Therapy/Photodynamic Therapeutic Agents for Malignant Melanoma

Abstract: A novel plasmonic gold nanocarrier using a modular RNA scaffold significantly improves delivery efficiency into diverse human cells, as presented by Norbert O. Reich and co-workers in article number 1602473. The orthogonal positioning of cell targeting peptides and functional RNA provides unprecedented control in the delivery of biologically active RNA. NIR light-triggered RNA release with spatio-temporal control further enhances the RNA delivery efficiency. Plasmon-Enhanced Spectroscopy Shell-isolated nanopar… Show more

“…Since melanomas can easily develop resistance to conventional chemotherapeutics, they are always associated with a very poor prognosis . Thus, the development of novel therapeutic methods for the treatment of malignant melanoma is a pressing need . Photothermal therapy (PTT) has been a promising therapeutic strategy for patients with melanomas due to its noninvasive, highly selective, and controllable treatment features .…”

“…The UV–vis absorption spectrum also showed the changes in the nanoparticles during the synthesis (Figure F). From the UV–vis absorption spectrum, it can be seen that, after coating the Fe 3 O 4 CNCs with PDA, the nanoparticles possessed a broad absorption in the NIR region; after grafting with mPEG and TPP, the absorption of the nanoparticles in the NIR region changed little, which is favorable for PTT . Considering the absorbance across the whole NIR region, Fe@PDA–TPP/SS could rapidly convert the light absorbed at NIR wavelengths into thermal energy …”

“…The quest for development of novel therapeutic strategies for treatment of melanoma led to preparation of polymerbased photo-thermal agents derived from poly-N-phenylglycine (PNPG), (prepared though N-phenylglycine (NPG) polymerization) for employment in NIR-mediated-phototherapy and generating ROS upon irradiation [56]. PNPG was further conjugated with HA as a targeting ligand and PEG-diamine as a potential coupling agent.…”
