Polyclonal activation of B lymphocytes during experimental infection withSchistosoma mansoni

Abstract: A significant polyclonal activation of B lymphocytes was observed during experimental infection of C57BL/10J mice with Schistosoma mansoni. The isotypic pattern of this expansion, assessed by the Protein-A plaque-forming cell method, was compared with and found to differ from those occurring after infection by Trypanosoma cruzi or injection of bacterial LPS. In the infection of S. mansoni an early expansion of most immunoglobulin isotypes occurs together with a late, sustained expansion of IgG1-secreting cells… Show more

“…polygyrus and igG 1 secretion. However, to our knowledge, C57BL/10, CBA and C3H mice are not intrinsically poor producers of IgGl to other antigens and parasites (Chapman et al 1979b, Lebrun & Spiegelberg 1987, Delgado & McLaren 1990, Lopes et al 1990. AKR mice were found to respond weakly with IgG1 relative to B 1 OBR mice following infection with Trichinella spiralis (Pond, Wassom & Hayes 1989) but were later reported to generate mainly IgG1 and IgM responses to group I1 antigens from the L1 stage of this parasite (Denkers et al 1990).…”

“…Preliminary experiments using CD4 ϩ T cell depletion and culture of purified B cells with SEAstimulated splenocyte supernatants indicate that FasL expression on B cells is dependent on a soluble factor from CD4 ϩ T cells (data not shown). This may explain why the percentages of FasL ϩ T and B cells are higher than the expected percentages of antigen-specific cells (15,23).…”

“…Splenomegaly is observed frequently in all stages of the disease, and it is characterized by the activation of germinative centers and enhancement of antibody production with polyclonal B cell activation. A robust, humoral response is initiated after 40 days of infection, reaching the highest levels of Ig production in the chronic phase of the disease [21,22]. The involvement of coelome-associated lymphoid tissues is also observed, and it is characterized by exacerbated macrophage reactivity and by extensive hyperplasia of myeloid and lymphoid lineages [23,24].…”
