Polygonatone H, a new homoisoflavanone with cytotoxicity from Polygonatum Cyrtonema Hua.

Abstract: A new homoisoflavonoid, (3R)-5,7-dihydroxy-6-methyl-3-(2'-hydroxy-4'-methoxybenzyl)-chroman-4-one (1), namely polygonatone H, in addition to fourteen known homoisoflavones (2-15) were isolated from the rhizome of Polygonatum Cyrtonema Hua. The structures were identified with the aid of 1D/2D NMR spectroscopic technologies. Compounds 2, 6, 8, 10, 11, 13, and 15 were isolated from P. Cyrtonema for the first time. Compound 1 showed cytotoxicities to human cancer cell lines with IC values to comparable those of ci… Show more

“…Also 5-OH (δ(H) 12.0) proton showed 3 J C-H correlations with carbon at δ(C) 96.0 (C-6) and 102.5 (C-11) which rule out the possibility of methyl attachment at (C-6) ( Figure 2). Based on this data and comparison with [18] compound 1 was assigned as 5,7dihydroxy-3-(4-methoxybenzyl)-8-methylchroman-4one which is reported for the first time to the best of our knowledge. Absolute configuration of 1 was assigned on basis of circular dichroism (CD).…”

“…By screening the differential quality markers between different origins of PR, it can be seen that peaks 9, 13, and 17 (VIP>1) play an essential role in grouping different origins of PR. LC/MS technology was used to analyze the retention time of the compound, and the mass‐to‐charge ratio fragment information, and matched with the information reported in the relevant literatures [59–61] . The three chemical marker components (9, 13 and 17) were identified as disporopsin, 5,7‐dihydroxy‐3‐(4′‐hydroxybenzyl)‐6,8‐dimethylchroman‐4‐one, (3R)‐5,7‐dihydroxy‐3‐(4′‐hydroxybenzyl)‐6‐methylchroman‐4‐one or isomer (Table 2, Figure S1–4).…”

“…Steroidal saponins are characterized by a skeleton derived from oxidosqualene, consisting of a sugar moiety linked to a steroidal (27 carbon atoms) aglycone [6]. Steroidal saponins have attracted extensive attention owing to their excellent anti-bacterial, anti-inflammatory, and anti-oxidation effect and the potential for alleviating diabetes and other metabolic disorders [5,7,8]. Steroidal saponins have been extensively used in the food, medicine, fodder, and cosmetics industries [9].…”
