Polymorphism rs4934 of SERPINA3 and Sporadic Intracranial Aneurysms in the Chinese Population

Abstract: Background: Serine protease inhibitor member 3 of clade A (SERPINA3) has been shown as a risk factor for aneurysmal subarachnoid hemorrhage in a Polish population. In the present study, the authors investigated the correlation between the rs4934 polymorphism of SERPINA3 and sporadic intracranial aneurysms in Chinese patients. Methods: The rs4934 polymorphism of SERPINA3 was identified by PCR and Taqman MGB probe analysis in genomic DNA from 275 patients with sporadic intracranial aneurysms (mean age 50.83 ± 10… Show more

“…A single nucleotide polymorphism (SNP) (rs4934) is located in the signal peptide region (at codon 15) in exon 2. It has been postulated that this genotype affects the expression of serpinA3 protein (12)(13)(14). Moreover, studies have proven that this genetic variant is a risk factor for aneurysmal subarachnoid hemorrhage in a Polish (12), but not in a Chinese (13) or Japanese (14) population.…”

“…We identified a subnetwork with SERPINA3 being the key molecule which is shown in Figure 3. SERPINA3 is known to be associated with several neurological disorders including Parkinson's disease [42], Alzheimer's disease [43], schizophrenia and cerebrovascular disease [44]. Notably, we observed GFAP and CHI3L1 which were upregulated in TBM to be part of this subnetwork.…”

“…Intracranial aneurysm (IA) occurs in $2% of the population worldwide, meaning that over 20 million people in China will experience an IA. 1 It is $1.6-times more common in women than in men, and the majority of ruptured IAs is observed in people between the ages of 40 and 60 years. 2 Rupture of an IA accounts for >90% of subarachnoid haemorrhage (SAH) cases.…”
