Polymorphisms in the interleukin-7 receptor α gene and mortality in untreated HIV-infected individuals

Abstract: The results suggest an association between the IL7Rα, rs6897932, T-allele and increased mortality among untreated HIV-infected, Zimbabwean individuals.

“…The presented data are in line with the previous study carried out in whites, although the primary aim differed (time to achieve CD4 þ T-cell count above 500 cells/ml and CD4 þ T-cell change) [15]. Likewise, we have found increased mortality of T-allele carriers in a Zimbabwean cohort of untreated individuals [23]. “…The genetic location of SNPs in IL7RA, whether coding or noncoding, and clinical associations of the SNPs are previously described [23]. To cover most common genetic variation within the IL7RA, we tagged the entire gene region by adding the following SNPs, rs11567762, rs1494565, rs10053847, rs11567714, and rs2229232,, resulting in a total of 10 tagging SNPs (R 2 ¼ 0.8, Haploview).…”

“…Apparently, our results of IL7RA polymorphisms appear to be discordant with previous studies, as these IL7RA SNPs have been previously related to AIDS progression in na€ ıve patients and CD4+ T-cell recovery in patients on cART [8][9][10]12,17]. However, we found IL7RA polymorphisms (rs6897932, rs987106 and rs10491434) were not associated with rapid AIDS progression in two Spanish large cohorts (CoRIS and LTNPs cohorts).…”

“…We selected three IL7RA polymorphisms located at three regulatory regions [rs6897932 (exon 6), rs987106 (intronic region) and rs10491434 (3 0 UTR)], which have been recently related to AIDS progression in ART-na€ ıve patients and CD4+ T-cell recovery in patients on cART [8][9][10]12,17].…”
