Polysialylation of NCAM Characterizes the Proliferation Period of Contractile Elements during Postnatal Development of the Epididymis

Abstract: Polysialic acid (polySia) attached to the neural cell adhesion molecule (NCAM) regulates inter alia the proliferation and differentiation via the interactions with neurotrophins. Since in postnatal epididymis neurotrophins and their receptors like the Low-Affinity Nerve Growth Factor Receptor p75 and TrK B receptor are expressed, we wanted to analyze if the polysialylation of NCAM is also involved during the development of the epididymis. To this end, we monitored the developmental changes in the expression of… Show more

“…In addition to scattered epithelial cells in all areas of the epididymis, primarily proliferating smooth muscle cells exhibit strong polySia staining directly after birth. Intriguingly, comparable results were also obtained, when the postnatal oviduct was examined [71]. In the epididymis during the first ten days after birth the amount of polysialylated NCAM decreases slightly, whereas a dramatic drop down was observed thereafter.…”

“…A comparable interrelation was also observed in the postnatal tunica albuginea [71], which enclose the testis and consists of fibroblast, myofibroblast and smooth muscle cells and extracellular matrix components like collagen [82]. An in cellulo study by Curtis and co-workers described a possible explanation for the contrarily-regulated amounts of polysialylated NCAM and collagen [83].…”

“…[49,50] To al esser extent, NCAM is expressed in skeletal myocytes,n atural killer cells,a sw ell as epididymal and pancreatic endocrine cells. [51][52][53][54] NCAM is known to modulate cell-cell interactions with counter-adhesion molecules by building homophilic and/or heterophilic interactions with other cell-adhesion molecules,f or example,t he neural cell adhesion molecule L1 or extracellular matrices. [55] As alarge "space-filling" cell-surface molecule,t he expression of poly-Sia reduces NCAM-mediated adhesive processes and NCAM-independent cell interactions.…”
