Positron Emission Tomography Imaging of the Gastrin-Releasing Peptide Receptor with a Novel Bombesin Analogue

Abstract: The gastrin-releasing peptide receptor (GRPR), a G protein-coupled receptor, is overexpressed in solid malignancies and particularly in prostate cancer. We synthesized a novel bombesin derivative, [68Ga]Ga-ProBOMB1, evaluated its pharmacokinetics and potential to image GRPR expression with positron emission tomography (PET), and compared it with [68Ga]Ga-NeoBOMB1. ProBOMB1 (DOTA-pABzA-DIG-d-Phe-Gln-Trp-Ala-Val-Gly-His-Leu-ψ(CH2N)-Pro-NH2) was synthesized by solid-phase peptide synthesis. The polyaminocarboxyla… Show more

“…was added to 25 μg of ProBOMB1 in 0.5 mL of sodium acetate buffer (0.1M; pH 4.5) and incubated at 95 C for 15 minutes. 15 To obtain the nonradioactive standard, NeoBOMB1 (3.0 mg, 1.9 μmol) and LuCl 3 (0.2M, 48 μL, 9.6 μmol) in 450 μL of sodium acetate buffer (0.1 M, pH 4.2) and 60 μL of MeCN were incubated at 90 C for 15 minutes and purified by HPLC using the preparative column (29.6% acetonitrile and 0.1% TFA in water; flow rate: 30 mL/min). Determination of molar activity was conducted using the analytical column (24% acetonitrile and 0.1% TFA in water; flow rate: 2.0 mL/min, retention time: 7.6 minutes).…”

“…ProBOMB1 ( Figure 1) was synthesized according to published procedures. 15 To obtain the nonradioactive standard, ProBOMB1 (1.46 mg, 88.7 μmol) and LuCl 3 (0.2M, 22.2 μL, 4.43 mmol) in 500 μL of sodium acetate buffer (0.1M, pH 4.5) were incubated at 95 C for 30 minutes and purified by HPLC using the semipreparative column (23% acetonitrile and 0.1% TFA in water; flow rate: 4.5 mL/min). was added to 25 μg of ProBOMB1 in 0.5 mL of sodium acetate buffer (0.1M; pH 4.5) and incubated at 95 C for 15 minutes.…”
