Post-therapeutic acute leukemia

Abstract: This study reports 35 cases of posttherapeutic acute leukemia and reviews the literature on this subject. These AL's are characterized by a high incidence of anemia, in particular refractory anemia, preceding the hematological disorder by several months, by the frequent finding of myelofibrosis, by the essentially granulocytic nature of the AL, and by the low rate of remission and the, in general, extremely short sruvival of a few months. These leukemias may develop following continuous chemotherapy with an al… Show more

“…We are indebted to the following physicians for providing information and slides ADDENDUM Since this manuscript was submitted for publication, several articles dealing with acute leukemia following prolonged cytotoxic agent therapy have been published (63)(64)(65). A 56-year-old white woman who was treated with radiation and combination chemotherapy (oyclophosphamide and adriamycin) for adenosquamous carcinoma of the endometrium, developed acute myelogenous leukemia 2% years later 963).…”

“…This patient received local radiotherapy and combination chemotherapy (imidazole carboxamide, vinblastine, melphalan and BCNU) for his solid tumor. Two additional patients with non-hematopoietic cancer, one with testicular seminoma and another with endometrial adenocarcinoma developed acute nonlymphoblastic leukemia 6 and 4 years later, respectively (65). Both patients received radiotherapy and one was also treated with combination chemotherapy, including alkylating agents.…”

“…In addition, it has been generally accepted that patients with secondary leukemia have a relatively poor prognosis. Early reports of patients with therapy-associated acute nonlymphocytic leukemia (t-ANLL) described median survival times of less than 3 months [1,9,10] and complete remission rates of only 10% [5,6,11]. The rising incidence of t-ANLL combined with its poor prognosis make this a serious problem of increasing magnitude.…”
